Study on the relationship between the expression of VEGFR2 and miR-27a-5p in gastric cancer tissues and clinicopathological parameters and prognosis

Abstract:

Objective

To study the relationship between the expression of VEGFR2 and miR-27a-5p in gastric cancer tissues, clinicopathological parameters and prognosis.

Methods

Data of 98 patients with gastric cancer who underwent laparoscopic total gastrectomy from January 2020 to December 2022 were retrospectively analyzed. Immunohistochemistry (IHC) and real-time quantitative fluorescent PCR (qRT-PCR) were used to detect the expression of VEGFR2 and miR-27a-5p mRNA in gastric cancer tissues and corresponding paracancer tissues (>5cm from the margin of cancer tissues). SPSS 20.0 software was used for statistical analysis. The expression level of miR-27a-5p and other quantitative data were expressed by () and independent sample t-test was performed. Clinical and pathological parameters and other statistical data were represented by [cases (%)], row Rank Sum test or χ2 test. Spearman Rank correlation was used to analyze the correlation between VEGFR2 and miR-27a-5p expression in gastric cancer tissues. The relationship with prognosis was analyzed by Kaplan-Meier method and Log-Rank test. P<0.05 indicates statistical difference.

Results

The high expression rate of VEGFR2 in gastric cancer tissues was significantly higher than that in para-cancer tissues, and the expression level of miR-27a-5p in gastric cancer tissues (1.08±0.42) was significantly lower than that in para-cancer tissues (2.51±0.85), with statistical significance (P<0.05), and the expression of the two was negatively correlated (r=-0.462, P=0.003). The high expression of VEGFR2 was significantly correlated with lesion diameter, T stage, differentiation degree, pTMN stage, vascular invasion, and lymph node metastasis (P<0.05), while the low expression of miR-27a-5p was significantly correlated with tumor differentiation degree, pTMN stage, vascular invasion, and lymph node metastasis (P<0.05). Kaplan-Meier analysis showed that the cumulative overall survival rate of the VEGFR2 high expression group was significantly lower than that of the VEGFR2 low expression group (81.0% vs. 94.7%) (P=0.047). The cumulative overall survival rate of miR-27a-5p expression group was significantly lower than that of miR-27a-5p expression group (81.7% vs. 92.6%) (P=0.041).

Conclusion

VEGFR2 is highly expressed in gastric cancer tissues, while miR-27a-5p is low expressed in gastric cancer tissues, and there may be interaction between the two, which is involved in the growth, proliferation, differentiation, invasion and migration of gastric cancer, thus affecting the course of gastric cancer, and is expected to become a potential target for diagnosis, treatment and prognosis evaluation of gastric cancer.
