Expression and clinical significance of Lnc RNA HIF1A-AS2 in colorectal cancer tissues

Abstract:

Objective

To investigate the expression of LncRNA HIF1A-AS2 in colorectal cancer tissues and to explore its clinical significance.

Methods

Clinicopathological data of 114 patients, who were with colorectal cancer from December 2012 to December 2015 and underwent radical resection, were collected, including samples of the cancer tissue and corresponding adjacent tissues (2 cm away from the lesion) and adjacent normal tissues (5 cm away from the edge of the cancer lesion). The relative expression levels of HIF1A-AS2 in colorectal cancer tissues, adjacent tissues and adjacent normal tissues were detected by qRT-PCR. The average expression level of HIF1A-AS2 in 114 enrolled patients was evaluated, and 89 patients with HIF1A-AS2 expression level greater than or equal to the average were divided into the high-expression group, while the rest were divided into the low-expression group. The relationship between HIF1A-AS2 expression and clinicopathological features was analyzed. Kaplan-meier survival curve was usedto analyze the 5-years survival of patients in two groups by using log-rank test. COX proportional risk regression model were used to analyze the independent risk factors for prognosis. A P value of < 0.05 was considered as statistically significant difference.

Results

The relative expression of HIF1A-AS2 in colorectal cancer tissues was higher than that in adjacent tissues and normal tissues (P<0.001). HIF1A-AS2 expression was significantly correlated with clinical stage and degree of differentiation in cancer tissue (P<0.05). The 5-year overall survival rate of the HIF1A-AS2 high-expression group was lower than that of the HIF1A-AS2 low-expression group (log-rank χ2=5.037, P=0.025). COX multi-factor analysis showed that lymph node metastasis, clinical stage Ⅳ, in poorly differentiated, with distant metastasis and HIF1A AS2 significantly high expression were the independent risk factors influencing the prognosis of patients with colorectal cancer (P<0.05).

Conclusion

HIF1A-AS2 is highly expressed in colorectal cancer tissues, which is closely related to clinicopathology and prognosis, and is one of the risk factors for poor prognosis of patients with colorectal cancer.
