The expression and clinical significance of Beclin-1 and SIRT-1 in Colorectal carcinoma

Abstract:

Objective

To investigate the expression of autophagy-related genes Beclin-1 and SIRT-1 in colorectal carcinoma, and to explore its clinical significance in the occurrence , development and prognosis of colorectal carcinoma.

Methods

The expression of Beclin-1 and SIRT-1 in 58 cases of colorectal carcinoma, 30 cases of colorectal adenomas and 30 cases of normal colorectal tissues were detected by using immunohistochemical methods, and the clinical and pathological characteristics were analyzed. Statistical analysis was carried out by using SPSS19.0 software. The counting data were tested by χ2 test. Spearman correlation analysis was used for correlation analysis. A P value <0.05 was considered as the statistical significant difference.

Results

The expression of Beclin-1 and SIRT-1 in colorectal cancer tissues (79.3%, 75.9%) were significantly higher than those in colorectal adenoma (53.3%, 50.0%) and normal colorectal mucosa tissues (20.0%, 23.3%) respectively with significant difference (all P<0.05). There was no correlation between Beclin-1 expression and clinicopathological factors in patients with colorectal cancer (P>0.05). The expression of SIRT-1 was positively correlated with the depth of invasion, lymph node metastasis and TNM stage of colorectal cancer (P<0.05). Beclin-1 expression was positively correlated with SIRT-1 expression (P<0.05). The 5-year survival rate of patients with positive expression of Beclin-1 and SIRT-1 was significantly lower than that of patients with negative expression of Beclin-1 (P<0.05).

Conclusions

Abnormal expression of autophagy-related genes Beclin-1 and SIRT-1 plays a synergistic role in the occurrence, development and prognosis of colorectal cancer.
