目的

研究胰腺癌血管生成样蛋白2(ANGPTL2)表达与免疫抑制细胞[髓系来源抑制细胞(MDSC)、调节性T细胞(Treg)、M2巨噬细胞]浸润数量的相关性,并分析ANGPTL2蛋白、MDSC、Treg和M2巨噬细胞与临床病理特征关系。

方法

收集2019年1月至2022年3月手术切除的胰腺癌组织标本及其癌旁正常组织(距癌灶边缘3~5 cm)标本各94例。以实时荧光定量PCR测定ANGPTL2 mRNA表达;免疫组织化学法检测免疫抑制细胞。采用统计学软件SPSS 25.0进行处理,ANGPTL2 mRNA表达等计量资料以(

±s)表示,采取配对样本t检验;癌组织ANGPTL2 mRNA表达及癌组织免疫抑制细胞浸润数量与病理特征关系分析,采用独立样本t检验或单因素方差分析,二者相关性分析采用Pearson相关性检验。P<0.05为差异有统计学意义。

结果

胰腺癌组织中ANGPTL2 mRNA表达水平及MDSC、Treg、M2巨噬细胞浸润数量较癌旁组织明显升高(P<0.05);胰腺癌组织中ANGPTL2 mRNA表达及免疫抑制细胞数量与分化程度、TNM分期、淋巴结转移等病理特征有关(P<0.05),二者呈正相关(r1=0.786,r2=0.752,r3=0.861,P均<0.05)。

结论

胰腺癌组织中ANGPTL2表达与免疫抑制细胞浸润数量均明显升高,且两者具有显著的正相关性,并与胰腺癌恶性程度及肿瘤转移有关。ANGPTL2可能通过调节免疫抑制细胞浸润,介导肿瘤细胞免疫逃逸,促进肿瘤细胞侵袭转移,从而诱导胰腺癌的发生发展。

Objective

To investigate the correlation between the expression of ANGPTL2(ANGPTL2)and the number of immunosuppressive cells(myeloid derived suppressor cells(MDSC),regulatory T cells(Treg),M2 macrophages)infiltrating pancreatic cancer,and analyze the relationship between AngpTL2 protein,MDSC,Treg and M2 macrophages and clinicopathologic features.

Methods

From January 2019 to March 2022,94 pancreatic cancer tissue specimens and adjacent normal tissue specimens(3~5 cm from the edge of the cancer focus)were collected. ANGPTL2 mRNA expression was determined by real-time quantitative PCR. Immunosuppressive cells were detected by immunohistochemistry. Statistical software SPSS 25.0 was used for processing,ANGPTL2 mRNA expression and other measurement data were expressed as(

±s),and paired sample t-test was adopted. Independent sample t test or One-way ANOVA were used to analyze the relationship between ANGPTL2 mRNA expression and the number of immunosuppressive cell infiltration in cancer tissues and pathological features,and Pearson correlation test was used to analyze the correlation between the two. P<0.05 was considered statistically significant.

Results

The mRNA expression level of ANGPTL2 and the infiltrating number of MDSC,Treg and M2 macrophages in pancreatic cancer tissues were significantly higher than those in paracancer tissues(P<0.05). The expression of ANGPTL2 mRNA and the number of immunosuppressive cells in pancreatic cancer tissues were correlated with the degree of differentiation,TNM stage,lymph node metastasis and other pathological features(P<0.05),and the two were positively correlated(r1=0.786,r2=0.752,r3=0.861,all P<0.05).

Conclusions

The expression of ANGPTL2 and the number of immunosuppressive cell infiltration in pancreatic cancer were significantly increased,and the two had a significant positive correlation,and correlated with the malignant degree of pancreatic cancer and tumor metastasis. ANGPTL2 may mediate the immune escape of tumor cells by regulating immunosuppressive cell infiltration,and promote the invasion and metastasis of tumor cells,thus inducing the occurrence and development of pancreatic cancer.
